National Museum of African American History and Culture Act of 2001 - Establishes within the Smithsonian Institution the National Museum of African American History and Culture to be operated as a center for scholarship and a location for museum training, public education, exhibits, and collection and study of items and materials relating to the life, art, history, and culture of African Americans form slavery and the era of reconstruction to the Harlem renaissance, the civil rights movement, and beyond.

Establishes the National Museum of African American History and Culture Council. Requires the Council to appoint a Director to manage the Museum. Authorizes the Council to plan, design, reconstruct, renovate, and expand the Arts and Industries Building to house the Museum.
